    Summary

    Pfam family PFAM:PF01877 {Protein of unknown function DUF54 } with 175 members in NR database and additional 89 members in the metagenomic datasets is represented in Archaea only.

    The first structural representative solved (PDB Id: TOPSAN:2nrq) was subject to FATCAT structural similarity search that indicated similarity to SCOP (v. 1.73) domain SCOP:d1iq4a_ classified as ribosomal protein L5 fold (RL5-like). However, no sequence similarity to this fold has been detected. Using 2nrq as query, DALI finds other DUF54 structures as hits with Z scores 15-10 like 2ogk, 3d7a, 2nwu or 2pzz. Next hit, with a score of Z=7.6, is with the 50S ribosomal protein L5 (1mji).

    2nrq protein structure corresponds to gene SSO0741 from S. solfataricus. Analysis of its genome context indicates a strong functional link (score 0.9) with the 50S ribosomal protein L15e (rpl15e gene).

    Based on fold similarity, one can hypothesize that this family is possibly a ribosomal protein and may bind nucleic acids [Ref].
