    Summary

    Pfam family PFAM:PF01949 {Protein of unknown function DUF99} with 52 members in NR database and additional 9 members in the metagenomic datasets is represented in Archaea, and Bacteria.

    The first structural representative solved (PDB Id: TOPSAN:2qh9) was subject to FATCAT structural similarity search that indicated similarity to SCOP (v. 1.73) domain SCOP:d1z6ra2 classified as ribonuclease H-like motif fold and superfamily. A DALI search using 2qh9 as query returns hits with endonuclease-V 3ga2, 3goc and 2w35 (Z=13) [Ref], followed by the argonaute protein structures 3hk2, 3f73 and 2f8s (Z=11) [Ref]. A Blast search with 2qh9 sequence (AF1433) returns a hit (e-val=1e-38; 29% seq. id.) with the endonuclease-V entry YP_001737819. Analysis of the genomic context of 2qh9 (gene AF1433) reveals a weak functional link (0.675) with rpoM gene that codes for a DNA-directed RNA polymerase subunit M.

    Based on marginal prediction from FFAS confirmed by structural similarity, one can argue that this family is remotely homologous to other families with the ribonuclease fold.
