    Protein Summary

    Caulobacter crescentus gene CC_0557 encodes the NP_419375 protein, a member of the PFAM ferritin clan, that has significant similarity to most members of this clan, but may eventually be classified as a separate family. There are about 30 full length sequence homologs with very broad phylogenetic distribution, from proteobacteria to Bacteroidetes to cyanobacteria. Much larger number of homologs are restricted to the first 250 residues with the most conserved sequence pattern limited to the first 80 residues. The "additional" 60 residues form a helical hairpin. Domains homologous to full lenght CC_0557 can also be found in large polyketide synthases (for instance in Salinispora arenicola VioB [YP_001537179]), suggesting that this is probably a part of a protein complex. The gene coding for CC_0557 protein is a member of a cluster of genes of unknown function, likely an operon. An unknown ligand is modelled at a highly conserved site (residues E21,H56, etc).

    Pre-SCOP classifies 3hl1 inside the ferritin-like superfamily. DALI search for structural similarity with 3hl1 as query,  confirms (Zscr=13) its homology to bacterioferritins (pdb ids: 1nfv, 2chp, 2vxi, 3hiu). The site for binding ions overlaps with the UNL location, and are similar to feritin (identical residues compared with 1nfv: E21, H56, H232, E121, E53, Y147, Y28 ). 

     

    Suggestion: co-xtal with Fe, and/or Fe-coproporphyrin iii (FEC) or maybe heme.
