    The gene YP_208451.1 from Neisseria gonorrhoeae encodes the N-terminal domain of  homologue of a human suppressor of fused protein (Sufu) PF05076.  The fold class of the protein is alpha and beta (a+b).

    There is only 1 significant hit in FFAS, to the human Suppresor of Fused (Sufu) protein (PDB id 1m1l, 15% sequence identity, superimposed in pink) which is involved in the Hedgehog signaling pathway. HHpred provides the same top hit, 1m1l, with an e-value of 7.3e-33, as well as 3D-Blast (e-val 7e-23). SSM gives a 53% of SSE match with 1m1l. FATCAT P-value for the same hit is 1.0e-07, and 16.7 for Dali Z-score.

    In humans, Sufu protein encodes a negative regulator of the Sonic Hedgehog signaling pathway.  The Hedgehog pathway is one of the primary signaling systems that specify patterns of cell growth and differentiation during vertebrate development.  The Hedgehog pathway drives proliferation and differentiation by activating the Gli/Ci family of zinc finger transcription factors. Gli/Ci proteins form Hedgehog signaling complexes with other signaling proteins, including the kinesin-like protein Costal-2, the serine-threonine kinase Fused (Fu), and Sufu.  Mutations in genes encoding components of this pathway are deleterious for normal development and are associated with cancer-predisposing syndromes (e.g. HOLOPROSENCEPHALY 3 (HPE3) [Ref];).  

     

    Corresponding pathways in bacteria are not known. But it is possible that this protein may be a bacterial evolutionary precursor of the mammalian Sufu. Alternatively, it may be involved in some different function that remains to be elucidated.

    Detailed structural, sequence and genomic context analysis have been peformed and will be added later and are in preparation for a manuscript.
